Advances on ferroptosis in hematological malignancies

摘要: 血液肿瘤是一类起源于造血系统和淋巴系统的异质性疾病,其核心特征在于恶性克隆性增殖与分化阻滞。临床常面临预后不佳与高复发风险的挑战。铁死亡是一种铁依赖性细胞死亡形式,由脂质过氧化驱动,现已被证实与血液肿瘤的发生、进展及治疗密切相关。系统综述铁死亡的分子机制,并重点探讨通过调控铁死亡相关基因与通路干预以白血病、多发性骨髓瘤、骨髓增生异常综合征、淋巴瘤为代表的血液肿瘤研究进展,以期为深入理解相关疾病进程及开发靶向治疗策略提供理论依据。

Abstract: Hematological malignancies are a group of heterogeneous diseases originating from the hematopoietic and lymphatic systems, characterized fundamentally by malignant clonal proliferation and differentiation arrest. Clinically, there are often significant challenges of poor prognosis and high risk of recurrence. Ferroptosis, an iron-dependent form of regulated cell death driven by lipid peroxidation, has been confirmed to be closely related to the pathogenesis, progression, and therapeutic response of hematological malignancies. This article systematically reviews the molecular mechanisms of ferroptosis. It focuses on the research developments of hematological malignancies represented by leukemia, multiple myeloma, myelodysplastic syndromes, and lymphoma through the regulation of ferroptosis-related genes and pathways in order to provide a theoretical foundation for a deeper understanding of the disease process and the development of targeted treatment strategies.
