G-四链体在抗肿瘤药物和神经退行性疾病方面的研究进展

The implication of G-quadruplex in cancers and neurodegenerative diseases

摘要: 转化医学是近年来医学领域中出现的一个新概念,旨在在基础研究和临床医疗之间建立更紧密的联系.富含鸟嘌呤(G)核苷酸序列形成的稳定G-四链体与人类重大疾病如肿瘤、神经退行性疾病密切相关,已成为转化医学研究中的一个重要方向.在端粒序列,G-四链体的形成和稳定可以抑制端粒酶活性,因此研发能稳定G-四链体结构的配体对抗肿瘤药物开发具有重大意义.此外,近年来神经退行性疾病中G-四链体的研究也取得重要进展.对与G-四链体相关的抗肿瘤药物研发和神经退行性疾病研究进行简要的总结.

Abstract: In the last decade, translational medicine came into the limelight, aiming to bridge the gap between basic research and clinical application. The association of G-quadruplex with life-threating and debilitating diseases is an emerging topic in translational medicine. Since the formation of G-quadruplex can inhibit telomerase activity, the ligands that can stabilize G-quadruplex have great significance for cancer therapeutics. In addition, growing evidence suggests the implication of quadruplex in neurodegenerative diseases. In this review, the research of G-quadruplex in the fields of anti-tumor drugs and neurodegenerative diseases is discussed.
