牛磺酸对小鼠腹腔巨噬细胞功能调节作用的体外研究

摘要: 目的 研究牛磺酸对小鼠腹腔巨噬细胞功能的调节作用。方法 收集小鼠腹腔巨噬细胞,调整细胞浓度为2×105/mL,分为5组,即正常对照组,LPS对照组(LPS终浓度为1 μg/mL)和低、中、高牛磺酸剂量组(牛磺酸的终浓度分别为5、50和500 μg/mL),培养24 h后ELISA法检测细胞上清中IL-12、TNF-α、IL-1、IL-6含量;瑞氏染色法检测并计算巨噬细胞对白色葡萄球菌的吞噬率和吞噬指数、Griess法检测吞噬葡萄球菌后细胞上清中NO的含量。结果 各剂量牛磺酸组的腹腔巨噬细胞TNF-α和IL-6含量均高于正常对照组(P<0.05),高剂量牛磺酸组IL-12的含量高于对照组(P<0.05),各剂量的牛磺酸组IL-1含量与对照组比较没有统计学意义;中、高剂量的牛磺酸组巨噬细胞对葡萄球菌的吞噬率、吞噬指数和上清中NO 的含量均高于正常对照组(P<0.05)。结论 牛磺酸可调节小鼠腹腔巨噬细胞IL-12、TNF-α、IL-6的分泌,但对IL-1的分泌没有调节作用,牛磺酸可促进巨噬细胞对葡萄球菌的吞噬和杀伤功能。

Abstract: Objective To observe the regulating effect of taurine on the function of peritoneal macrophages of mice in vitro. Methods Mouse peritoneal macrophages were collected and the cell concentration was adjusted to 2 × 105/mL. The mouse peritoneal macrophages were divided into 5 groups,which were normal control group, LPS control(LPS final concentration 1 μg/mL), 3 dose of taurine groups(final concentration of taurine was 5 μg/mL, 50 μg/mL and 500 μg/mL, respectively). The IL-12, TNF-α, IL-1, IL-6 in macrophages supernatant were determined by ELISA. The phagocyte rate and phagocytic index of macrophages on Staphylococcus Albus were detected by Whight's stain and the content of NO in the supernatant of after phagocytosis was measured by Griess. Results The levels of TNF-α and IL-6 in peritoneal macrophages in every taurine group were higher than those in normal control group(P<0.05). The content of IL-12 in the high dose taurine group was higher than that in the control group(P<0.05). There was no significant difference in IL-1 content between the taurine group and the control group. The phagocytic rate, phagocytic index and supernatant NO of macrophages in the medium and high dose taurine group were higher than those in the normal control group(P<0.05). Conclusion Taurine can regulate the secretion of IL-12, TNF- α, IL-6 in peritoneal macrophages of mice, but does not regulate the secretion of IL-1, and taurine can promote the phagocytosis and killing function of macrophages to staphylococcus.
