旋毛虫寄生致小鼠十二指肠组织细胞凋亡

Study on apoptosis of duodenum of mice infected with Trichinella spiralis

摘要: 目的 观察旋毛虫实验感染小鼠后不同时间成虫寄生处十二指肠组织细胞凋亡的情况。方法 建立旋毛虫实验感染小 鼠动物模型,取其感染 3、5、7 d 成虫期小鼠十二指肠组织常规切片,HE 染色法观察组织病理变化,TUNEL 法检测十二 指肠细胞凋亡指数。Western-blot 方法检测十二指肠组织中 Caspase-3 表达水平变化,对旋毛虫成虫致小鼠十二指肠组织 细胞凋亡情况进行综合评价。 结果 HE 染色结果显示,随着感染时间的延长,十二指肠组织的病理损伤加重。TUNEL 结 果显示,小鼠十二指肠组织凋亡细胞指数随着感染时间延长明显增多(P<0.05),Western-Blot 方法分析 Caspase-3 表达 量随着十二指肠组织感染时间的延长而增加(P<0.05)。 结论 旋毛虫成虫可诱导小鼠十二指肠组织细胞凋亡,而和感染 时间呈正相关。

Abstract: Objective To study the apoptosis of duodenum of mice infected with Trichinella spiralis MethodsInfection model was established, duodenum tissues of the mice were taken in the time of 3, 5, 7days. The histopathological changes were observed, TUNEL method was applied to detect the duodenum apoptotic index, Western-Blot method was used to detect caspase-3 expression level changes in the duodenum tissue of mice of different infection periods. The apoptosis state induced mouse duodenum tissue was conducted comprehensive evaluation. Results HE staining showed that as the infection time went on, the pathological damages of the duodenum were increased TUNEL results showed that the index of apoptotic cells of the mouse duodenum tissue changed with infection time increased significantly(P<0.05), Western-Blot analysis indicated that Caspase-3 expression levels increased with the duodenum tissue prolonged infection time(P<0.05). Conclusion Adult Trichinella spiralis can induce apoptosis in duodenum tissue of the miceand the time of infection was positively correlated.
