Abstract: Objective To study the effects of different solvents (sterile water and PBS buffer) on the polypeptide antigen’s emulsification. Methods Dissolved polypeptide antigen was completely emulsified with complete Freund’s adjuvant by the way of pull-and-push with an artificial blender connecting three-way pipe and two yringes. The effects of emulsified mixture were evaluated by water drip test. Results The emulsification time was longer and the procedure was easy to control by taking the sterile water as solvent. The stable water in oil emulsifier was formed in a short time by taking the PBS buffer as solvent, whereas it was easy to get over emulsified. Conclusion The effects of different solvents on the polypeptide antigen’s emulsification were different significantly.
