Effects on ultrastructure of neuron and blood brain barrier in ischemia reperfusion by pretreatment with 3-n-butylphthalide in rats

Abstract: ObjectiveTo observe the effects on ultrastructure of neuron and blood brain barrier (BBB) in ischemia reperfusion by pretreatment with 3-n-butylphthalide in rats. Methods The 30 SD rats were divided randomly into five groups: sham group, ischemia reperfusion group, NBP different dose groups (NBP 20 mg/kg group, NBP 40 mg/kg group, NBP 80 mg/kg group), NBP groups were respectively administrated with NBP 20, 40, 80 mg/(kg·d) one week, the ischemia reperfusion models were made, and then evaluated the neurological deficit score of the rats, the permeability of blood brain barrier were detected by content of Evans Blue (EB) in the brain tissue of ischemic reperfusion. At the same time, The ultrastructure of neuron and blood brain barrier were observed in brain tissue with electron microscope. Results The results showed that the neurological deficit scores and content of Evans Blue (EB) in NBP groups were obviously decreased compared with ischemia reperfusion group(P<0.01, P<0.05) and the ultrastructural injury of neuron and blood brain barrier of the brain tissue were improved with ischemic reperfusion in rats. Conclusion 3-n-butylphthalide pretreatment can exert protective functions on ultrastructure of neuron and blood brain barrier in the cerebral ischemia reperfusion injury.
