Relationship between fragile sites gene of WWOX in typeⅡpneumocytes and idiopathic pulmonary fibrosis

Abstract: Objective To investigate changes in expression of WWOX, a fragile sites gene, in typeⅡpneumocytes isolated from rats with idiopathic pulmonary fibrosis. Methods SD rats were randomly divided into 3 groups (n=6 in each group):sham-operated group, model group and tetramethylpyrazine group. After given bleomycin, the rats were harvested at 7、14 and 28 day to isolate the typeⅡpneumocytes and then culture the cells in vitro. Then, we detected the protein expression of WWOX in pneumocytes with immunocytochemistry. Results Compared with the sham-operated group, no significant differences in WWOX protein expression were found in rat typeⅡpneumocytes of the model group administrated with bleomycin for 7 days. After 14 and 28 days of administration, the WWOX protein expression was reduced greatly in the model group. Nevertheless, tetramethylpyrazine enhanced significantly WWOX protein expression compared with the model group. Conclusion In this study, we demonstrate that the expression of fragile sites gene of WWOX is altered during idiopathic pulmonary fibrosis, while tetramethylpyrazine suppresses the alteration to some extent, suggesting the medication could play a protective role for idiopathic pulmonary fibrosis.
