Effects ofα-zeranol on the IL-6 expression and secretion in rat osteoblasts

Abstract

Abstract: Objective To investigate the effects ofα-zeranol on the IL-6 expression and secretion in rat osteoblasts. Methods Osteoblasts were isolated from the skull of SD neonatal rats and identified by morphological observation, ALP staining, and collagen I immunolfuorescence staining. 17β-estradiol andα-zeranol with different concentration were administrated. The expression of IL-6 mRNA in the osteoblasts was detected by RT-PCR and the level of IL-6 in culture supernatant of osteoblasts was measured by ELISA. Results α-zeranol with 10-10~10-5 mol/L concentrations could inhibit the expression and secretion of IL-6 in osteoblasts, among which the concentrations of 10-6 and 10-7 mol/L had obvious effects. Conclusionα-zeranol can inhibit the expressions of IL-6 mRNA and protein in osteoblasts, it may play an important role in the prevention of postmenopausal osteoporosis.
