    Effects ofα-zeranol on the IL-6 expression and secretion in rat osteoblasts

    Objective To investigate the effects ofα-zeranol on the IL-6 expression and secretion in rat osteoblasts. Methods Osteoblasts were isolated from the skull of SD neonatal rats and identified by morphological observation, ALP staining, and collagen I immunolfuorescence staining. 17β-estradiol andα-zeranol with different concentration were administrated. The expression of IL-6 mRNA in the osteoblasts was detected by RT-PCR and the level of IL-6 in culture supernatant of osteoblasts was measured by ELISA. Results α-zeranol with 10-10~10-5 mol/L concentrations could inhibit the expression and secretion of IL-6 in osteoblasts, among which the concentrations of 10-6 and 10-7 mol/L had obvious effects. Conclusionα-zeranol can inhibit the expressions of IL-6 mRNA and protein in osteoblasts, it may play an important role in the prevention of postmenopausal osteoporosis.

    Objective To explore the inlfuence of sperm membrane ultrastructure damage on the non-obstructive infertile male. Methods After embedding the fresh semen from one case of infertile male, using transmission electron microscopy (TEM) to observe the sperm plasma membrane, outer acrosomal membrane , inner acrosomal membrane etc. Results Under the electron microscope, the defects of plasma membrane, outer acrosomal membrane and inner acrosomal membrane in the semen sperm of this infertile man were examined, with some of other abnomal spermultrastructures observed, such as defects in the sperm acrosome, nucleus, neck, mitochondrial and microtubules. Conclusion The non-obstructive male infertility is closely related to the abnormality of sperm membrane ultrastructure.

    Objective Survivin gene is the strongest apoptosis-inhibition factor among inhibitor of apoptosis protein (IAP), and purification of fusion protein of functional domains (baculovirus IAP repeat, BIR) may provide the foundation for interaction research between the gene and related other genes. Methods BIR sequence, after ampliifcation with PCR and validation by Escherichia coli DH5αthrough pGEX4T-2 vector, was transformed into E. coli BL21. Fusion protein of BIR was induced by IPTG and puriifed by Glutathione Agarose Affinity Chromatography. Results The amplified fragment BIR was about 276bp, purified fusion protein molecular weight of approximately 37ku. Conclusion PCR results are consistent with the expected fragment size, recombinant plasmids after 2h after IPTG induction, the cell in the fusion protein increase signiifcantly, which provide basis for further research on Survivin gene function and its interacting protein.

    Objective To explore the changes of serum amyloid A (SAA), plasma D-dimer and tumer marker CA724 for predicting tumor metastasis of patients with colorectal cancer. Methods Preoperative SAA, D-dimer and CA724 were measured in 58 patients with colorectal and 55 patients with benign disease, and they were analyzed with statistics method. In addition, according to pathological TNM (AJCC/UICC TNM), the patients with colorectal were divided into no metastasis group, regional lymph node metastasis group and distant metastasis group for comparing SAA, D-dimer and CA724 between groups. Results SAA, D-dimer and CA724 levels were signiifcantly higher in patients with colorectal carcinoma than those with benign diseases (P<0.05). SAA test showed that there was statistical significance between no metastasis group, regional lymph node metastasis group and distant metastasis group, and there was no statistical signiifcance by CA724. SAA and D-dimer levels in distant metastasis group were obviously higher than that of no metastasis group and regional lymph node metastasis group (P<0.05). Conclusion There is guiding signiifcance for conifrming regional lymph node metastasis and distant metastasis in patients with colorectal to detect SAA CA724 and D-dimer levels.

    Objective To study the relationship between expression of PTEN and survival prognosis of NSCLC. Methods 62 cases with non-small cell lung cancer conducted radical surgical resection and 14 cases done adjacent normal tissue were selected and examined PTEN protein by immunohistochemical SP method to explore its relationship with the survival of the patients. Results The results indicated that there were statistically significant differences between PTEN expression of cancer and normal tissues adjacent groups. There were statistically signiifcant differences about the PTEN expression between the positive group and the negative group (P<0.05). Conclusion The low expression of PTEN in non-small cell lung cancer shows that PTEN plays an important role in the development of non-small cell lung cancer, which is signiifcantly associated with the survival time of the patients.

    Objective To investigate expression of osteopontin (OPN) and adhesion molecule CD44v6 in colon adenocarcinoma and their clinical signiifcance. Methods 80 cases of colon cancer tissue were detected by immunohistochemical methods (with lymph node metastasis in 32 cases, 48 cases without lymph node metastasis), 30 cases of colorectal adenoma, 20 cases from colon tumor (polyp, adenoma, cancer, etc.) 5 cm above normal tissue osteopontin and CD44v6 expression levels were examined to analyze the correlation of osteopontin and CD44v6 expression in colon adenocarcinoma. Results Adenocarcinoma of the colon and surrounding lymph nodes, osteopontin and CD44v6 expression were signiifcantly higher than that of normal colon tissue and colonic adenoma expression levels (P<0.05), colon adenocarcinoma with metastasis in osteopontin CD44v6 protein expression levels were significantly higher than that of non-metastatic colon adenocarcinoma (P<0.05). Colon adenocarcinoma osteopontin expression and expression of CD44v6 were in a state of consistency (P<0.05). Conclusion OPN-CD44v6 with colon adenocarcinoma, developing consistent, osteopontin and CD44v6 joint detection for colon cancer development and prognosis explicitly provide a basis for clinical diagnosis and treatment , which has a certain signiifcance in clinical practices.

    Objective To get the clinical comparative observation on outcomes of antituberculosis fixed-dose combination and plate combination drug with initial tuberculosis treatment, providing a scientiifc basis for the antituberculosis fixed-dose combination clinical application. Methods Patients with initial treatment tuberculosis were divided into treatment group and the control group. The treatment group was treated with anti tuberculosis ifxed-dose combination, while the control group was treated with plate combination drug. Two groups all used the alternate day therapy program (2H3R3Z3E3/4H3R3). Observe the treatment completion, loss, program change and discontinuation of the medicine of the two groups with the method of control study. Results For the tuberculosis patients of initial treatment smear-positive and initial treatment smear-negative, the treatment completion rate of antituberculosis ifxed-dose combination group was greater than that of plate combination drug group(P<0.05). Loss rate of antituberculosis fixed-dose combination group was less than that of plate combination drug group(P<0.05). Program change rate of antituberculosis ifxed-dose combination group was less than that of plate combination drug group(P<0.05). The rate and the frequency of discontinuation of the medicine of antituberculosis ifxed-dose combination group was less than that of plate combination drug group(P<0.05). Conclusion Antituberculosis fixed-dose combination is advantageous to help medical staff on patient therapy management and supervision, and the application of it in the National Tuberculosis control programme is feasible.

    Objective A retrospective analysis in terms of clinical characteristics and detection was carried out for a case of Enterobacter cloacae bearing NDM-1 isolated from a urine specimen of an in-patient in our hospital in July 2012. The purpose of this paper was to render suggestions for controlling the epidemic of“superbugs”based on discussions regarding its antibiotic-resistant characterization and mechanism. Methods According to the recommendations of CLSI, susceptibility testing for antibiotics using K-B and MIC methods was performed. Carbapenemase production and its phenotypic were confirmed by modified Hodge test and metal-enzyme test. PCR for detecting and sequencing the antibiotic-resistance gene was also performed. Results This isolate was resistant to 15 antibiotics such as imipenem, meropenem and others which were classiifed into 7 categories. This isolation showed positive for modiifed Hodge test and screening test for detecting metal-enzyme phenotype. The DNA sequence of the isolation amplified by PCR showed 99%similarities comparing with NDM-1 gene cited in Genbank. It was conifrmed as the ifrst report of E. cloacae carrying NDM-1 by the CDC of Heibei Province.Conclusion The NDM-1-producing isolation may be related to the elder patient who was inifrm, catheterized and experienced multi-drug treatments. Therefore, in order to lessen the incidents of multi-drug resistant strain, it is ideal that we should focus on the control over catheterization, the use of antibiotics and taking strict sterilization.

    Objective To observe the clinical curative effects of manual diaplasis and small splint immobilization combined with rehabilitation guide in treatment of Colles fracture. Methods 68 cases with stable Colles fracture were treated with manual diaplasis, small splint immobilization and Shi’ Jiegudan, and given rehabilitation guide. The clinical curative effect of 3 months was appraised. Results All patients were followed up for 4-10 months with an average of 6 months. All of them achieved bony union and clinical healing standard. Clinical healing time was 21-39 days with an average of (28.02±1.64) days. 65 cases were cured, 3 cases improved. The cure rate was 95.59%, and the total efifciency was 100%. Conclusion The rehabilitation guide of the patients treated with small splint immobilization is effective clinically. It is worthy for being widely used in clinical orthopedics.

    Objective To observe the effects of routine rehabilitation training combined with medium-frequency electrotherapy in the treatment of patients with shoulder-hand syndrome after stroke. Methods 84 cases with shoulder-hand syndrome after stroke were randomly divided into the treatment group ( 42 cases )and a control group ( 42 cases). Two groups were given rehabilitation training for 4 weeks, and the treatment group received medium-frequency electrotherapy meanwhile. Upper limb motor functions, activities of daily living and upper extremity pain and swelling were measured apart by the Fugl-Meyer assessment, MBI index, and visual analog scores. The indicators before and after treatment were compared. Results There were 80 patients completed the trial (including treatment group 40 cases, control group 40 cases), and two group patients’ upper limb pain, edema, FMA and MBI scores compared with those before the treatment were signiifcantly improved (P<0.05), the treatment group improved more signiifcantly than that of the control group (P<0.05), the efifciency of the treatment group and the total efifciency was better than that of the control group (P<0.05). Conclusion Routine rehabilitation training combined with medium-frequency electrotherapy is a good method for the treatment of shoulder hand syndrome after stroke.
    Resecting large primary dumbbell tumor tangled with abdominal organs in lumbar region through single-stage posterior approach

    Objective To evaluate the feasibility, safety and effect of resection large spinal-retroperitoneal dumbbell tumor tangled with vital abdominal organs through independent posterior approach. Methods 18 patients' clinical data, operation records, pathologic examination results and follow-up data were included in this retrospective analysis. In the initial 8 patients, combined surgical approaches from back and front were used. The later 10 patients were operated on using an independent improved posterior approach to remove tumor. Results In earlier 8 patients who underwent a combined surgery from posterior and antero-lateral approaches, total surgical time averaged 5 hours, the blood loss averaged 800 mL. 1 patient died of brain metastasis 5 months post-surgically. 7 cases symptoms disappeared, no tumor recurrence. In later 10 patients, only independent posterior approach was used, operative time averaged 3 hours, blood loss averaged 450 mL. All patients were not tumor recurrence and symptoms disappear. Conclusion Primary large dumbbell tumor in lumbar region which tangles with abdominal organ can be safely removed through independent posterior approach. Compared with combined posterior and anterior approaches, independent posterior approach is more simple, less traumatic, and similarly effective.

    Objective To observe the effects of dialectical applying ear acupuncture point buried beans to reduce the pain in justification of common type of primary dysmenorrheal. Methods Fifty cases of patients with primary dysmenorrhea in after syndrome differentiation were given corresponding ear acupuncture point buried beans. 0.5 h later pain reliefs with patients were assessed and the recurrence of the patients with periodic pain was examined. Results In iffty patients with various types of primary dysmenorrheal, 49 cases eased the pain in half an hour, the total effective rate was 98.0%. In 36 patients with periodic pain, 31 cases relieved, the effective rate was 86.1%. Conclusion The method of dialectical applying ear acupuncture point buried beans can effectively alleviate pain in patients with primary dysmenorrhea.

    Objective To evaluate the effects of chlorine-containing disinfectant on TB sputum preprocessed by pancreatic enzyme. Methods 116 samples from sputum smear-positive pulmonary tuberculosis patients collected from outpatient service were randomly divided into the control group and treatment group. 58 samples of TB sputum in treatment group were preprocessed by 5%pancreatic enzyme disinfector, then to 84 disinfector of containing 5 000 mg/L chlorine. The 58 samples in control group, were directly processed by 84 disinfector of containing 5 000 mg/L chlorine. After 0.5 h, 1 h and 2 h of the treatment, the positive rates of mycobacterium tuberculosis of each sputum samples were comparatively analyzed. Results The positive rates of mycobacterium tuberculosis of 0.5 h, 1 h and 2 h in control group were 100%, 88%and 100%respectively. After pretreatment of pancreatic enzyme, the positive rates of 0.5 h, 1 h and 2 h in treatment group were 62%, 14%and 0 respectively.Conclusion The pretreatment of pancreatic enzyme on sputum of pulmonary tuberculosis patients can obviously promote the disinfectant effects. It should be actively promoted in disinfectants on TB sputum.

    Objective To study the diversity between medical students with internet addiction and the control and analyze the effecst of internet addiction. Methods 687 medical students were conducted with retrospective investigation on the situation of internet use over the past semester. Results The rates of internet addiction and internet dependence were 4.8% and 8.3% respectively. The academic record, skipping classes depended on internet, net play in classes, preferences of after-school activities and the purpose of net play had signiifcantly differences between internet addiction group and the controls(P<0.05). Conclusion Internet addiction has a negative impact on study.

    Objective To evaluate the effects of fast walking exercise on the physical activity in elderly patients with type 2 diabetes (T2DM) in glucose metabolism and physical activity indicators. Methods 80 patients with T2DM were randomly divided into two groups:exercise group and the control group, 40 cases in each group. Exercise group performed on 16 weeks of fast walking, before the movement of glucose metabolism and physical activity indicators were compared. Results The experimental group iftness program VO2max before and after exercise, 30s arm lfexion and extension, 2min standing still, chair-reach, grab the back stretch, 6min walk, six test scores were signiifcantly increased (P<0.05). While continuous improvement 30s standing and sitting in the chair stand test results go two (P<0.01). In the control group it had no signiifcant improvement after the test. Patients in the control group iftness body each index changed little (P>0.05). 2h postprandial blood glucose (2hPG) decreased significantly (P<0.05), glycosylated hemoglobin (HbAlc), postprandial 24h urine positive rate, fasting urine positive rate was signiifcantly lower (P<0.01) fasting plasma glucose (FPG), fasting insulin (Flns) and 2h postprandial insulin (2hIns) levels did not change signiifcantly (P>0.05). Lower extremity muscle strength tests comparing exercise group had ankle flexion and extension muscle strength growth to varying. Conclusion Fast walking exercise training program will help to improve glucose metabolism and activity in patients with T2DM.

    Objective To investigate how spouse synchronization of health education affects the subjective well-being of the female patients with urinary incontinence. Methods 106 cases with urinary incontinence of healthy spouse were randomly divided into two groups. In observation group 53 cases of patients and their spouses simultaneous implementation of health education. In the control group, 53 cases were only received health education to herself. Results Before the education, the difference between Health Education Social Support Rating Scale for four groups of patients and SWB was no statistically signiifcance (P>0.05). After conducting spouse synchronization of health education one month, 3 months, the different indicators and SWB scores of the experimental group were higher than that of the control group (P<0.05). Conclusion Through conducting the spouse synchronous of health education, the social supports can be signiifcantly increased, and it is also helpful for the patients to get quick recovery.

    Some single Chinese herb can be used for the prevention and treatment of pulmonary ifbrosis.First of all, this paper describes the mechanism of pulmonary fibrosis in cell and molecular level. Secondly, this paper describes single Chinese herb prevention and treatment of pulmonary fibrosis experiment progress in cell and molecular level to ifnd the prevention and treatment of pulmonary ifbrosis drug targets as a theoretical basis for clinical treatment.

    Objective To explore anchored instruction strategy in the application of general nursing practice teaching and evaluation. Methods The three-year higher vocational nursing students as investigating object were investigated, anchored instruction strategy based on constructivism learning theory to general nursing training classes for the teaching design and application designed teaching effect after implementation of the questionnaire were conducted. Results 94.1%of the students considered that the application of this teaching strategy was helpful to stimulate the learning interest, and cultivate the information search, analysis and solving problems abilities, and the ability of cooperation to help others, but also shortened the gap between theory and practice. Conclusion The anchored instruction strategy applied in general nursing training courses has signiifcant difference in cultivating good information literacy and innovation ability of nursing students.

    Objective To study the advantages and theoretical basis of merging the two courses ---Child and Adolescent Health(CAH), Child Care(CC). Methods Comparative analysis was used to compare the contents of the course syllabus and teaching plans of CAH and CC with the public health physician examination syllabus. Results The results showed that most of the contents of CAH and CC were repeated, as the same knowledge of this two courses was taught respectively, more class hours were spent. This study found that the same knowledge of the two courses still play important role after the two courses had been merged. In addition they conformed to the contents of the public health physician examination syllabus. Conclusion Repeating the same contents and wasting class hours could be avoided by merging the two courses, the connections of this two courses could be enhanced by this way, contents of merging conform to the public health physician examination syllabus.

    Objective To investigate the role of PBL teaching method in clinical practice of thoracic oncological surgery. Methods 34 students of clinical medical specialty in Xinjiang Medical University were randomly divided into PBL group (Problem-based learning group, n=17) and (traditional LBL group traditional lecture-based learning group, n=17). The effectiveness of these two different teaching group were compared. Results Both average scores of common knowledge and clinical case analysis in PBL group were signiifcantly higher than those of LBL group. The student's ability of self learning, the way of clinical thinking and the capability of knowledge understanding in PBL group were signiifcantly better than those in LBL group. Conclusion The way of PBL can improve the effectiveness of medical student's learning in the clinical practice of thoracic oncological surgery, enhance their capability of patients treatment. The PBL teaching method in practice of thoracic oncological surgery has certain promotion value.
