C-erbB-2、Bmi-1 及PTEN 基因在大肠癌组织中的表达及意义

摘要: 目的 研究C-erbB-2、Bmi-1 及PTEN 基因在大肠癌组织中的表达及其临床意义。方法 选取50 例大肠癌标本及50 例癌旁正常大肠黏膜组织进行免疫组化检测,并分析C-erbB-2、Bmi-1 及PTEN 基因在两者中的表达及三种基因之间相关性研究。结果(1)C-erbB-2、Bmi-1 基因在大肠癌组织中的表达均明显高于在正常大肠组织中的表达,PTEN 基因在大肠癌组织中表达显著低于在正常大肠组织中的表达,差异有统计学意义。(2)C-erbB-2、Bmi-1 基因与大肠癌的淋巴转移、分化程度、浸润深度、Ducks 分期呈正相关,其与大肠癌的发生、发展及生物学行为具有促进作用;而PTEN 基因与大肠癌的淋巴转移、分化程度、浸润深度、Ducks 分期呈负相关,与大肠癌的发生、发展及生物学行为具有抑制作用。(3)大肠癌组织中C-erbB-2、Bmi-1 及PTEN 基因之间具有相关性,其中C-erbB-2、Bmi-1 基因在大肠癌的发生发展过程中成正相关;C-erbB-2、PTEN 基因在大肠癌的发生发展过程中呈负相关;Bmi-1、PTEN 基因在大肠癌的发生发展中呈负相关。结论 C-erbB-2、Bmi-1 和PTEN 基因在大肠癌发生发展中起一定作用,联合检测可能成为预后、靶向治疗的标志物。

Abstract: Objective To study expression of C-erbB-2, Bmi-1 and PTEN gene in colorectal carcinoma and its clinical significance. Methods 50 cases of colorectal carcinoma tissues and 50 cases of adjacent normalcolorectal mucosa were detected by immunohistochemical method, and the expression of C-erbB-2, Bmi-1 and PTEN gene in the study of the correlations and three genes were examined. Results (1) The expression of C-erbB-2, Bmi-1 gene in colorectal cancer tissues were significantly higher than the expression in normal colorectal tissues the expression of PTEN gene in colorectal carcinoma was significantly lower than the expression in normal colorectal tissues, the difference was statistically significant. (2) C-erbB-2, Bmi-1 gene in colorectal cancer and lymph node metastasis, degree of differentiation, depth of invasion, Ducks stage were positively correlated, and the incidence of colorectal cancer, the development and biological behavior had a role. While PTEN gene in colorectal cancer and lymph node metastasis, degree of differentiation, depth of invasion, Ducks staging were negatively related with colorectal cancer, the occurrence, development and biological behavior of inhibitory effect. (3) There was a correlation between C-erbB-2, Bmi-1 and PTEN genes in 3 colorectal cancer tissues, where C-erbB-2 and Bmi-1 genes in the development of colorectal cancer was related to the occurrence and development process. C-erbB-2 and PTEN genes of colorectal carcinoma was negatively correlated. Bmi-1 and PTEN gene in colorectal cancer development was negatively correlated. Conclusion C-erbB-2, Bmi-1 and PTEN gene in the occurrence play a role in the development of colorectal cancer, combined detection may be a prognostic marker and targeted therapy.
