黄独乙素对胃癌细胞株 SGC-7901 增殖的影响及其机制

Effects of Disobulbin B on proliferation of human gastric carcinoma cell line SGC-7901

摘要: 目的 研究黄独乙素对人胃癌细胞 SGC-7901 增殖的影响及相关的作用机制。方法 用 RPMI-1640 培养液对 SGC-7901 细胞进行传代培养,用不同浓度(0~160μg/mL)黄独乙素分别对 SGC-7901 细胞进行处理,应用 CCK-8 法检测细胞的增殖率;光学显微镜观察细胞形态学改变;Western blot 检测细胞周期蛋白 CyclinD1 表达的变化。结果 黄独乙素以计量依赖方法抑制胃癌细胞生长,抑制率最高达 57.26%; 细胞形态发生明显改变;CyclinD1 蛋白水平表达显著下调。结论 黄独乙素能显著抑制 SGC-7901 细胞的增殖,其机制可能与下调细胞周期蛋白 CyclinD1 的表达有关。

Abstract: Objective To investigate effects of Disobulbin B (DB) on the proliferation of human gastric carcinoma cell line SGC-7901 and explore the possible mechanism. Methods SGC-7901 cells were treated with different concentration (0~160 μg/mL) of DB. Cell proliferation was detected by CCK-8 assay; cell morphology changes were observed under optical microscopy; The expression of CyclinD1 was detected by Western blot. Results DB inhibited the growth of gastric cancer cells in a does-dependent manner and the highest inhibition rate was 57.26%.The structure of SGC-7901 cells changed greatly. The protein expression level of cyclinD1 was reduced. The effects were increased with the increase of the DB concentration. Conclusion DB could inhibit cell proliferation in SGC-7901 cells by down-regulating the expression of CyclinD1.
