结直肠癌与抑癌基因SLC5A8甲基化的相关性

摘要： 目的探讨结直肠癌组织中SLC5A8基因mRNA转录水平及其甲基化状态。方法分别采用实时荧光定量PCR法及甲基化特异性PCR法(MSP)检测40例结直肠癌组织、癌旁组织SLC5A8基因mRNA转录水平及其甲基化水平。结果 SLC5A8基因在结直肠癌组织中mRNA转录水平显著低于癌旁组织(χ²=16.241,P<0.05);结直肠癌组织中SLC5A8基因甲基化阳性率显著高于癌旁组织(χ²=32.237,P<0.05);该基因的mRNA转录水平与其甲基化状态呈负相关,相关系数为0.875。结论 SLC5A8基因甲基化能显著降低该基因的表达,与结直肠癌的发生可能存在一定关联。

Abstract: Objective To investigate SLC5A8 gene’s transcription level of mRNA and its methylation status in colorectal cancer.Methods Real-time fluorescence quantitative PCR and methylation specific PCR method(MSP)were used to detect the transcription level of SLC5A8 gene’s mRNA and the methylation level of SLC5A8 gene in 40 cases of colorectal cancer tissues and adjacent tissues.Results The rate of transcription level of SLC5A8 gene’s mRNA in colorectal cancer rate was significantly lower than that in tumor adjacent tissues(χ²=16.241,P<0.05),the positive rate of SLC5A8 gene methylation in colorectal cancer tissues was significantly higher than that in adjacent tissues(χ²=32.237,P<0.05),the transcription level of this gene’s mRNA was negatively correlated with its methylation status.The correlation coefficient was 0.875.Conclusion SLC5A8 gene methylation can significantly reduce the expression of the gene,which may be associated with the occurrence of colorectal cancer.
