Abstract: Objective To explore the methylation and transcription level of SLC5A8 gene in breast cancer. Methods The tumor samples and adjacent normal tissue samples of 32 cases of breast cancer patients treated with basic surgery in Affiliated Hospital of Hebei University from May 2017 to December 2019 were divided into two groups, namely the experimental group and the control group. Methylation of SLC5A8 gene in 32 cases of breast cancer and surrounding normal tissues was detected with methylation-specific PCR, and transcriptional expression of SLC5A8 gene in 32 cases of breast cancer was detected with RT-PCR. Results There were 20(62.50%)positive cases of SLC5A8 methylation in 32 breast cancer tissues in the experimental group and 7(21.87%)positive cases of SLC5A8 methylation in 32 adjacent tissues in the control group. The methylation positive rate in the experimental group was significantly higher than that in the control group(P<0.05). mRNA transcription was detected in 6 of the 20 breast cancer patients(30.00%). mRNA transcription was detected in 7(58.33%)of the 12 cancer tissues with negative methylation in the experimental group. In the experimental group, the transcription expression rate of methylated positive oncotissue was significantly lower than that of methylated negative oncotissue(P<0.05). Conclusion SLC5A8 gene methylation is a common phenomenon in breast cancer, which may be involved in the occurrence and development of breast cancer, and The methylation of SLC5A8 gene in breast cancer tissues is not completely consistent with the transcriptional expression of the gene, and there is a certain inverse consistency between the two.
